Tesofensine Tracker

Track tesofensine — the appetite-suppressing compound

Tesofensine is an oral triple monoamine reuptake inhibitor (serotonin, noradrenaline, dopamine) originally studied for neurological disease and later found to cause significant weight loss — about 10% in a Phase 2 trial. It is investigational and not FDA approved for obesity. Unlike GLP-1 drugs, it is a small molecule, not a peptide. Shotlee is the free app to track your protocol.

01BACKGROUND

What is tesofensine?

Tesofensine is an oral small molecule that blocks the reuptake of three neurotransmitters — serotonin, noradrenaline, and dopamine — which suppresses appetite and increases energy expenditure. It was first investigated for Parkinson’s and Alzheimer’s disease, where researchers noticed marked weight loss as a side effect.

In the Phase 2 TIPO-1 trial, tesofensine produced roughly 10% mean weight loss at 24 weeks — about double what other agents achieved at the time. It is being developed further (including for rare hypothalamic obesity) but is not FDA approved for general weight management.

⚠️
Investigational — not a peptide
Tesofensine is an investigational small-molecule drug, not a GLP-1 peptide, and is not FDA approved for obesity. Grey-market tesofensine is unregulated. This is educational; decisions belong with a clinician.

Protocol FAQs

Tesofensine is an oral triple monoamine reuptake inhibitor (serotonin, noradrenaline, dopamine) studied for weight loss after it caused marked weight loss in neurological-disease trials. It produced about 10% weight loss in Phase 2. It is investigational, not FDA approved.

In the Phase 2 TIPO-1 trial, tesofensine 0.5 mg averaged about 10% mean weight loss at 24 weeks — roughly double comparators at the time. Results in further development are still being characterized.

No. Tesofensine is a small-molecule drug that works on brain neurotransmitters, not a GLP-1 peptide. It is a different mechanism entirely from semaglutide or tirzepatide.

No. It is investigational and not FDA approved for obesity. Grey-market tesofensine is unregulated research material.
