GLP-1 Pen Clicks Calculator
Convert pen clicks to your dose
On a standard U-100 multi-dose pen, each click of the dial is 1 unit, which equals 0.01 mL. So your dose in clicks is the same as your dose in syringe units — for example, 5 mg of tirzepatide from a 10 mg/mL pen is 50 units, or 50 clicks. Use the charts below, and always confirm against your device and prescriber.
How pen clicks map to your dose
On most U-100 multi-dose injection pens, each click of the dial delivers 1 unit, and 1 unit is 0.01 mL. That means clicks and U-100 syringe units are the same number — so your dose in clicks equals (dose ÷ concentration) × 100, exactly like the units math.
For example, with a pen filled at 10 mg/mL, a 5 mg dose is 0.5 mL = 50 units = 50 clicks; a 2.5 mg dose is 25 clicks. The higher your concentration, the fewer clicks per milligram.

How to count clicks safely
Accuracy & safety
Click-counting is only as accurate as your pen’s increment and your concentration. A miscounted concentration or click size changes the real dose, so double-check both, and prefer a pen with a visible dose window when possible.
This tool is educational and does the clicks-to-dose math only. It does not endorse off-label or compounded use; follow your prescriber and your device’s instructions, and confirm doses with your pharmacist.
Frequently Asked Questions
On a U-100 pen where 1 click = 1 unit, a 5 mg dose from a 10 mg/mL pen is 50 clicks (0.5 mL). From a 20 mg/mL pen it is 25 clicks. Clicks equal units = (dose ÷ concentration) × 100.
Branded single-dose Zepbound and Mounjaro pens are pre-set to a fixed dose and are not click-adjustable. Click-counting applies to refillable/multi-dose U-100 pens, such as those used for compounded tirzepatide.
On most U-100 pens, yes — 1 click = 1 unit = 0.01 mL. Some pens step in 0.5-unit or 2-unit clicks, so check your device before counting.
Multiply clicks by 0.01 mL (for a 1-unit pen) to get volume, then multiply by your concentration in mg/mL. Example: 50 clicks = 0.5 mL; at 10 mg/mL that is 5 mg.
Yes, for a standard 1-unit-per-click U-100 pen the clicks number equals the syringe-units number. If your pen uses a different increment, adjust accordingly.
